All About Human Mesenchymal Stem Cells

Mesenchymal stem cells (MSCs) originate from the mesoderm during embryonic development and give rise to multiple different tissue types including bone, cartilage, and fat. It is important to note that while the embryonic mesenchyme produces both connective tissue and hematopoietic tissue types, MSCs do not differentiate into hematopoietic cells.

MSCs are typically isolated from bone marrow, however, they can also be obtained from other tissues including blood (peripheral or cord), placenta, fat, and muscle. One particularly rich source of MSCs is the umbilical cord. Also known as Wharton’s Jelly, umbilical cord tissue has a higher concentration of MSCs than the umbilical cord blood and has the advantage of being an easily obtainable tissue source.

Human MSCs are a robust stem cell culture model:

  • Under optimized cell culture medium conditions, MSCs can divide (or self-renew) in vitro while maintaining their multipotent stem cell state.
  • On the other hand, MSCs can easily differentiate into cell types of the mesenchymal lineage including osteocytes, myocytes, adipocytes, and chondrocytes, when subjected to the appropriate differentiation culture conditions.

MSCs provide ample opportunity to conduct studies on basic biology, development, stem cell biology, stem cell differentiation, and cell fate.

Differentiated cell types derived from normal human MSCs also provide a healthy source of cells or tissue that can be used to study various debilitating diseases such as osteoporosis (bone loss), osteoarthritis (cartilage loss), sarcopenia (muscle wasting), obesity, diabetes, inflammatory diseases, and cancer.

Lifeline® HMSCs in the Latest Research

Numerous researchers rely on our high quality normal human MSCs, optimized MSC media, and differentiation kits for their scientific studies. Here are a few highlights from recent publications:

  • Talukdar et al. used Lifeline® normal human MSCs derived from lipospirate and bone marrow along with StemLife MSC medium to look at the effects of different 2-D graphene nanoparticles on MSC viability and differentiation ability. They determined which types of nanoparticles were tolerated by MSCs and did not alter their differentiation potential. This study suggests that specific graphene nanoparticles can be used to label MSCs and their cell derivatives for imaging and therapeutic purposes.
  • A separate study by Prugh et al. looked at the role of acetylcholinesterase (AChE) in bone marrow-derived MSC proliferation and differentiation. Using Lifeline® cell staining kits, they were able to characterize differentiated cell types derived from bone marrow MSCs including chondrocytes (Alcian blue), osteoblasts (Alizarin Red), and adipocytes (Oil Red O). They found that MSCs exposed to organophosphates had reduced AChE activity and were unable to differentiate efficiently into adipocytes.
